Effects of Different Concentration of Vitreous on Cultured Bovine Retinal Pigment Epithelial Cells ; In Vitro Study


Abstract
Proliferative vitreoretinopathy(PVR) is characterized as the growth of cells in the vitreous gel and on the surface of the retina in eyes with complicated retinal detachment. Based on morphological studies, it has been postulated that retinal
pigment
epithelial(RPE) and glial cells migrate into the vitreous where they proliferate, acquire characteristics of myofibroblasts, synthesize an extracel lular matrix and contract.
In recent years it has been clear that the extracellular matrix plays an active role n regulating behvior of cells that come in contact it, The formed vitreous gel is a specialized extracellular matrix composed mainly of collagen and hyalouronic
acid.
RPE cells are normally protected from exposure to vitreous by the overlying retina. There is a high incidence of PVR in eyes with giant tears.
However, the factors controlling RPE cells behavior in PVR are poorly understood. To investigate the effects of vitreous on RPE cells¢¥, we treated cultured bovine RPE cells with different concentration of vitreous and its effects on cell growth
were
determined. We also examined the possiblility of the change of vitreal compoent between pre-and post-filtration by polyacrylamide gel electrophoresis. Bovine RPE cells were counted with coulter counter.
@ES The results are as follows :
@EN 1. The whole vitreous stimulated the proliferation of bovine RPE cells. Proliferation of RPE cells treated with 10 %, 20 %, 40 % of vitreous were more significantly increased than in controls(P<0.05) and reached to maximum at 20 %
concentration.
2. Microscopic examination of vitreous treated bovine RPE cells showed no morphologic characteristics and changes of growth pattern.
3. Electrophoresis indicated no changes of vitreous compoent between pre-and post-filtration of vitreous and maximal band was found at protein with molecular weight 66 KD.
These results suggest that vitreous components stimulate the proliferation of RPE cells and the vitreous gel network only affect themorphological characteristics of RPE cells.
